Kemampuan Whole Cell Helicobacter pylori dalam Menginduksi Degradasi Kolagen Tipe IV Melalui Peningkatan Aktivitas Makrofag

Abstract

ABSTRAK

Helicobacter pylori  berperan dalam infark miokard  akut dan penyakit jantung koroner.  Sitotoksin H.pylori  dapat menginduksi sitokin inflamasi IL1-β dan TNFα yang mengaktivasi enzim proteolitik MMP-9 dan menyebabkan degradasi kolagen melalui plak aterosklerosis. Penelitian ini dilakukan untuk membuktikan kemampuan whole cell H.pylori dalam menginduksi degadrasi kolagen tipe IV melalui peningkatan aktifitas makrofag. Aktivitas makrofag dilihat dari kadar TNFα dan IL1-β yang diukur dengan metode Elisa. Hasil uji ANOVA mengindikasikan perbedaan signifikan (p<0,001) aktifitas makrofag pada kelompok normal dan whole cell H.pylori. Produksi enzim MMP-9 dapat dideteksi melalui gelatine zymography dilanjutkan dengan western blotting pada berat molekul 93 kDa. Fragmenliasi kolagen didapatkan pada fragmen 61,2 kDa  dan 29 kDa. Dapat disimpulkan bahwa H.pylori dapat menginduksi degradasi kolagen tipe IV melalui peningkatan makrofag. Temuan ini juga mendukung patomekanisme infark miokard.Kata Kunci : Kolagen Tipe IV, makrofag, whole cell H.pylori
